The dramatic loss of function seen with trimer-preferring bNAbs against L193A and I423A, two mutations in a core-proximal position at considerable distance from the V2-glycan and V3-glycan epitopes, reinforces the concept that global allosteric effects rather than epitope disruptions are responsible for the altered neutralization profiles.
