Fetal brains were obtained from 3 pregnant rats at pregnant day 17. Primary cortical neurons were isolated and pooled separately from each mother and plated onto 100mm ´ dishes, for a total of 4 plates from each pregnant rat. At Day in Vitro (DIV) 16, 3 of the 4 platesfrom the same mother, were exposed to either 1 or 3 MAC halothane (about 1% or 3%) or 3 MAC isoflurane (about 4%) in a gas tight chamber inside the culture incubator for 6 hr. The 4th plate from each mother treated with air were used (inside the same incubator, but outside the chamber) as controls. Isoflurane at 1 MAC was not conducted because of the complete lack of transcriptional response in the animals exposed to 1 MAC of isoflurane. The cells were recovered for an additional 6 hr, harvested, frozen by liquid nitrogen, and stored at –80oC. |
